Abstract
Study on the effect of caffeine within egg yolk Tris semen extender to the quality of separation of Garut ram sperm has been conducted. The aims of the study were to know the effect of caffeine in egg yolk Tris extender on motility and velocity of the sperm after separation and to find out the optimal dose of caffeine for both parameters. This study used Completely Randomized Design with four treatments on different dose of caffeine (0 mM; 2 mM; 4 mM and 6 mM) and each treatment was repeated five times. Results showed that addition of caffeine into egg yolk Tris extender was significantly affect sperm motility (P<0.01) of Garut ram sperm after separation and the best dose of caffeine was 4 mM to be added into Egg Yolk Tris semen extender to obtain the highest sperm motility and velocity.
Rezumat
S-a organizat un studiu privind efectul cafeinei adaugate în diluantul pe baza de albus de ou Tris asupra calitatii spermei provenita de la berbeci Garut. Scopul a fost acela de a investiga efectul adaosului de cafeina în diluantul Tris cu galbenus de ou asupra mobilitatii si vitezei de înaintare a spermatozoizilor, respectiv stabilirea dozei optime de cafeina pentru a afecta pozitiv ambii parametri. S-a utilizat un design experimental complet randomizat, cu 4 tratamente ce au presupus dozari diferite ale cafeinei (0 mM; 2 mM; 4 mM si 6 mM), fiecare tratament fiind repetat de 5 ori. Rezultatele au aratat ca adaosul de cafeina în diluantul Tris pe baza de galbenus a afectat semnificativ motilitatea spermatica (P<0.01) a materialului seminal de la berbeci Garut dupa dilutie. Cel mai bun dozaj al cafeinei a fost de 4 mM în diluant, pentru a obtine motilitate si viteze maxime ale spermatozoizilor.
